HTRF® Transcreener™ ADP Assay The New Solution for ADP DetectionPresenter Francoise Degorce, Cisbio

The assay is a competitive immunoassay in which native ADP and d2-labeled ADP compete for binding to an anti-ADP MAb labeled with Eu(K). Enzyme-free detection allows for low false positive rates. This homogeneous non-radioactive solution is well suited for high-throughput kinase and ATPase screening applications. Here, we show the performances of the HTRF® Transcreener™ ADP assay on enzymes producing ADP. Signal stability, S/B and Z’ factor were determined on a mixture of constant concentrations of ATP and ADP in order to mimic the conversion of ATP into ADP. Finally, a simple protocol for enzyme assay development was validated using reference inhibitors for IC50 determinations (enzyme titration, enzymatic kinetics, Km).
