Rapid Generation of Epitope-Specific Recombinant Antibodies Using in vitro Selection TechnologyPresenter Andreas Boll, MorphoSys AG, Germany
Additional Authors: Christian Frisch Recombinant antibodies selected from vast combinatorial libraries have created a new paradigm in monoclonal antibody generation. High affinity monoclonal antibodies against virtually any antigen can be isolated, expressed and purified in many formats from E. coli in massive throughput in about 8 weeks. The well-established HuCAL GOLD® library is the source of many antibodies being developed as human therapeutics by leading Pharma companies, and is now available for generation of monoclonal antibodies for research. Examples will be presented which highlight the differences between the in vitro generation of recombinant antibodies and traditional animal immunization approaches (i) a plethora of engineering possibilities including a choice of tags, labels and fusions to functional domains, (ii) optimized antibody formats for special applications like protein arrays, (iii) targeting of specific epitopes both positively and negatively by guided selection and (iv) generation of sandwich pairs by selection on antibody/antigen complex.
