Automation & Miniaturization of Immunoassays for Drug Discovery  AlphaLISA®, a Sensitive No-Wash Assay

AlphaLISA®, an immunoassay method for the detection of various analytes and biomarkers, offers many advantages over ELISA-like technologies. AlphaLISA assays are homogenous, requiring not a single wash step. Samples can be tested directly with no dilution step due to the very large dynamic range of the assays. Based on two beads and a chemical energy transfer, assays are simple to develop and rapid to perform. Assays are very sensitive, allowing working with small sample volumes (1-5 µL). AlphaLISA assays are therefore easy to miniaturize and automate, enabling efficient High-throughput Screening.Results showing quantitative detection of insulin in various 96-, 384- and 1536-well microplates will be presented. Assays were performed in 10 to 50 µL assay volumes in only 90 minutes. Lower Detection Limits (LDL), dynamic ranges and signal to background ratios for each plate format will be compared. The insulin and other biomarker assays were fully automated on the JANUS® Automated Workstation, with very low variation and assay parameters similar to assays performed in manual mode. Excellent sensitivities were obtained, demonstrating how easy and simple miniaturization and automation are using AlphaLISA. AlphaLISA is the easiest ELISA ever.
