Liquid chromatography is well-established as a valuable tool for carrying out separation-based biological assays by its ability to separate substrate and product prior to detection and analysis. However, continued assay miniaturization has resulted in smaller and smaller sample volumes, testing the limits of conventional LC approaches. Microscale liquid chromatography (microflow LC), requiring only small amounts of sample (~1uL injection volumes and femtograms of sample consumed per LC run), is well-suited for this application. Recent advances have resulted in the commercialization of microflow LC systems optimized for speed, performance and reliability. Combined with sensitive microfabricated absorbance and fluorescence detection systems, microflow LC offers a flexible and robust platform that provides fast, reliable results for minizaturized assays. We will present the results of assays carried out using the Eksigent ExpressLC microflow LC system, including both absorbance and fluorescence detection data. We will discuss the benefits of using microscale and nanoscale technologies for separation-based biological assays.
