Immobilized Bead Assay System For Multiple Analyte Detection

We have developed a multiparameter detection system for the simultaneous detection of up to 18 analytes in small volumes. This system is based on color-coded microbeads immobilized within a microplate. The beads are coated with capture antibodies and immersed in serum, cell lysate or supernatant solutions. The homogeneous assay principle is realized as a competitive binding assay on the surface of the microspheres. Using an automatic multi-color analysis of the individual microplate wells with an automated microscope individual beads are classified according to their antibody coating. The number of bound analyte molecules is detected simultaneously. The system is fully compatible with all common microplate liquid and plate handling systems, and is used to determine the IC50 value of IgG, IgM, and IgA in a competitive model binding assay. The binding kinetics of these immunoglobulins are analyzed by repeated measurements of a single class of microspheres.
